I’m often asked about developments in this market, but this year, 2011, has seen one real trend – outsourcing. Big pharma and biotech are always looking to develop their pipelines, but what has struck me is that some pharmaceutical companies in the UK are going about it in a whole different way.

Pharma-Biotech Collaboration

Attracting smaller companies to collaborate is one way of identifying novel, high-value platforms and targets to fill the pipeline. Big companies such as Glaxo SmithKline (UK) are creating a large complex around their facilities in Stevenage (Stevenage Biosciences Catalyst) to foster this type of relationship. There doesn’t seem to be shortage of innovative biotech companies with late-stage drugs that are looking for a sales and marketing partner. This is where the expertise of the established pharmaceuticals comes into play, their established network means that these new drugs are easy to get onto the market.

Big Pharma Turning to Bio-generics: the next big thing in 2012

Looking ahead, I’m predicting that the production of bio-generics will take a big leap. In this market the big players will also be the companies that currently produce their proprietary biologicals. One way of protecting their interest will be the production and sales of their off-patent compounds as generics. That is, when the patent runs out on a licensed drug, the same pharmaceutical company will produce the generic. This ensures these companies a considerable market share, since their branded products are known and proven on the market, as are their production processes and marketing channels.

Nowadays there are multiple methods to generate monoclonal antibodies (i.e. hybridoma, B-cell selection and cloning, phage display, transgenic animals etc). The main issue is to define the antibody product profile before you embark on an antibody generation process.

1.	Think about what the antibody has to do

Firstly you need to think about the profile of the antibody, consider what it is that you really need it for. What characteristics does the antibody have to have in regard to affinity, isotype, effector functions, cross reactivity, biological efficacy, size, and stability for example?

Monoclonal antibodies are useful tools for both medical treatment, diagnostics as well as research. Taking this into account, the application will define the specification and critical parameters. These issues can be dealt with during the process of antibody generation, optimisation and production in a QBD (Quality By Design) approach using a combination of currently available (recombinant) antibody technologies.

2.	Determine the limiting factor for your specific approach

Is it time, money or something else? Only when you understand this can you determine the best approach for your budget and purpose.

3.	Finally, find the right service provider that can integrate and coordinate all your requirements to generate the required antibody molecule

Ensure that the antibody service provider has an integrated platform of various technologies and methods that are required to generate and deliver the antibody you require.

Effective Rheumatoid Arthritis Antibody Treatment Identified From Company’s Antibody Generation Technologies and Mouse Modelling, Showcasing At Bayer

22 November 2011: ModiQuest Research, Nijmegen, The Netherlands: Jos Raats, CEO of ModiQuest has been invited to Bayer, Germany on 22 November to give a seminar on their novel, effective antibody-based treatments for Rheumatoid Arthritis.

The discoveries were made as a result of ModiQuest Research’s state-of-art proprietary technology platforms for efficient antibody generation, antibody cloning and reformatting, stable cell line development and antibody production facilities. These were used in combination with the company’s own highly effective rheumatoid arthritis mouse model, to identify and characterise promising antibody preclinical candidates. First and second generation mouse and human monoclonal antibodies have been identified and characterised as being able to slow or halt the onset and progression of arthritis in preclinical studies.

Together with early diagnostic testing which is already on the market, the discoveries by ModiQuest herald new possibilities for rheumatoid arthritis treatment that can be applied before the onset of clinical symptoms and irreversible joint damage. For established rheumatoid arthritis, the group demonstrated that these new antibodies have the potential to significantly decrease arthritis symptoms, in combination with current anti-inflammatory treatments.

Technologies For Developing New Antibody Treatments Showcasing At Human Antibodies and Hybridomas (HAH) 2011

25 October 2011: ModiQuest Research, Nijmegen, The Netherlands: Combining accurate, early diagnostic tests for autoimmune disease, and new antibody treatment options are two themes being presented by antibody technology experts, ModiQuest Research at HAH 2011 (7-9 November 2011, Cannes, France). ModiQuest Research’s technology includes monoclonal antibody generation and specialist immunological reagents to support the research and development of new antibody therapies and diagnostics. President and CEO, Dr Jos Raats will present latest data on new antibodies and early diagnostics for rheumatoid arthritis, using their hybridoma and phage display platforms.

Dr Raats said, “The Human Antibodies and Hybridomas event will be a great opportunity to share knowledge with others in the field of antibody development. I am honoured to be delivering a paper entitled ‘Anti-Citrulline Antibody as Novel Therapeutic Drug in early Rheumatoid Arthritis,’ detailing ModiQuest Research’s development of a novel therapy for Rheumatoid Arthritis (RA). Using our antibody generation technologies, this new drug candidate was developed by focussing on targets like Deiminated Peptide-Epitopes (DPE), since a number of studies indicated that these deimination modifications could be responsible for the initial triggering of autoimmunity and the breaking of tolerance. We use the same technologies to support our clients in the development of new antibody therapies and diagnostics. ”

Modiquest Research are proud to be sponsors of Human Antibodies & Hybridomas 2011. This year the scientific programme will include keynote lectures, contributed papers and poster presentations emphasising all key aspects of human antibody and hybridoma technology, applications and engineering. The scope of the conference falls broadly into eight main session categories; infectious diseases, autoimmunity, molecular biology, emerging technologies, clinical applications, therapeutic antibodies, cancer and anti-infective antibodies.

ModiQuest Research offers an intelligent approach to antibody generation, cloning and cell line production. Its offering includes monoclonal antibody generation, cell line services, plus antibody cloning and sequencing. Products available in the range include immunological reagents to enable early research into new antibody therapeutics and diagnostics. These products contain unique and highly specialised reagents to enable research in autoimmune disease, infectious diseases including fungal and parasitic diseases and oncology and are available to purchase online via ModiQuest’s website.
