Phage Display

The strength of our phage display platform lies in our range of proprietary human libraries and our ability to develop custom-made phage libraries from different species. As a result, we can rapidly and efficiently develop highly specific antibodies that meet the particular needs of preclinical research studies.

Human Antibodies for Clinical Studies

Our human phage display library approach is an effective route for developing therapeutic antibodies for clinical studies.  We have a range of human libraries from healthy individuals and patients, which typically have higher levels of specific antibodies against certain immune disease antigens.

Custom Phage Display Library Generation

We can immunize several animal species, even using a mix of antigens, and generate a custom antigen-biased immune library, allowing the selection of in vivo matured high affinity antibodies with the desired characteristics. This approach maximizes the chances of obtaining highly specific antibodies with high affinity.

For example, for rapid generation of high affinity antibodies, we can immunize chickens with the target antigen and generate a target-specific phage display library within 4-6 weeks.  Subsequent phage selection using the target antigen usually yields antibodies with high specificity and high affinity, making them useful for preclinical studies.

Low Immunogenic Targets

Phage display is also a good choice for low immunogenic antigens, including rare targets and difficult epitopes.  Using a combination of B-cell selection and phage display, we can first enrich for the target antigen-specific antibody producing cells and then clone the antibody cDNAs encoding the target specific antibodies.  This enables us to generate monoclonal antibodies from different species. It also allows us to compete out particular undesirable antibody features to increase specificity if required.

Difficult to Purify/Produce Antigens

Some clients come to us with antigens that are difficult to produce or purify for further immunization work.  In such cases, we have successfully generated monoclonal antibodies by DNA immunization, using DNA clones expressing the antigen of interest.

Typical Phage Display Process, Deliverables and Time Lines

Step Process Deliverables & Time Line
Library prep & QC Initial generation of a target-specific recombinant antibody library, or QC and preparation of our in-house proprietary libraries 3-8 weeks (new library) or 2 weeks (Modiquest Research Library)
Selection of recombinant target antibodies We perform on average 4 selection rounds by panning the phage library against one target. 2-3 weeks
Analysis and characterization of selected recombinant antibodies ELISA-based screening and fingerprint analysis of target specific clones. Up to 5 positive antibody cDNA clones delivered in 4-6 weeks
Follow-on options Sequencing of positive antibodies
Affinity maturation
Production and purification of scFv antibodies
Recloning of scFv antibodies in human, mouse or chicken full-size immunoglobulins and transient production in CHO or HEK293 cells
Stable cell line generation
Timeline is dependent on clients’ specific needs.
