Collagen-induced arthritis is an antibody mediated disease. Therefore, type II collagen specific antibodies can be used to induce arthritis in mice. Our mixture of type II collagen specific antibodies (2.6mg per animal) induces severe inflammation in the ankle and paws of DBA mice which resemble many aspects of arthritis. One of the biggest advantages of our monoclonal antibody-induced arthritis model is that severe arthritis is induced within 24-48 hours instead of the four weeks required to induce arthritis after immunization with type II collagen. ModiQuest Research recommends highly susceptible DBA/1 mice. Other strains may be used, however the amount of antibody cocktail required may vary. Thus, this model is ideal for rapidly screening therapeutics for the treatment of arthritis and to study the mechanisms involved in the development of arthritis. |

PAD enzymes are transcriptional co-regulators that catalyze the calcium dependent conversion of specific arginine residues present in for instance histones to citrulline. In diseases such as multiple sclerosis and rheumatoid arthritis (RA) these PAD enzymes seem to be dysregulated. For instance in RA these enzymes become active when they exit the cell in regions of joint inflammation where they deiminate arginines in proteins such as vimentin and fibrinogen. The level of PAD activity has been correlated with RA severity. The ABAP test is also suitable for determining PAD activity of purified recombinant PAD enzymes used in various bioassays.

The MQR-ABAP test is a powerful method to rapidly screen for PAD activity. Compared to currently used assays for PAD activity determination such as the DAMO BAEE conversion test the MQR-ABAP assay is more robust, and at least 500 times more sensitive. ModiQuest Research can generate monoclonal recombinant antibodies in completely customized procedures and for client specified antigens.
All recombinant monoclonal antibodies can be expressed in bacteria as scFv or Fab, or as full size fully human, fully mouse, fully chicken antibody formats, or in any chimeric antibody form required by the client, in mammalian cells.
Both transient expression (up to 10 liter scale) and stable expression systems are provided. High producing stable cell lines can be efficiently generated using our automated stable cell line generation procedures.

ModiQuest Research and Pepscan announced today the start of a research collaboration to identify human and animal derived "me-too" monoclonal antibodies and monoclonal antibodies against intractable disease targets. These targets include membrane proteins like GPCRs and ion channels.
Using its proprietary CLIPS™ technology Pepscan will generate functional mimics of intractable targets. ModiQuest will apply various methods like proprietary B-cell selection and cloning, or phage display to generate potent functional antibodies against mimics of the essential parts of such intractable targets.

Hybridoma generation
ModiQuest Research offers high-throughput generation of hybridoma cell lines. Our team uses an in-house developed highly efficient electrofusion technology. This unique technology is linked to the ClonePix-FL technology to perform high-throughput automated screening and collection of antigen specific clones.
This innovative approach:
Using this approach ModiQuest Research can screen large numbers of hybridomas in a high-throughput manner, hereby improving the success rate and production levels of the selected hybridomas.
Electrofusion
By combining our highly efficient proprietary electrofusion method with the ClonePix-FL technology, our team can rapidly generate high quality mouse and rat hybridomas. An outline of the approach and time lines is as follows: ModiQuest Research guarantees to deliver positive clones, using its standard protocols.
B-cell selection and electrofusion
For difficult or poorly immunogenic antigens we recommend our antigen B-cell selection technology. This technology allows for the selection of rare antigen-specific B-cells from immunised animals or humans with an active B-cell response.
Using mice, the selected B-cells are subsequently used to generate hybridomas. For other species (rabbit, lama, and human), the antibody-encoding cDNA from antigen-specific B-cells is cloned and expressed in our proprietary recombinant antibody expression system. This approach can be used for: As such, this approach has a higher efficiency than standard hybridoma generation.
Phage display
ModiQuest Research has extensive experience in phage display technology. Using our phage display methods in combination with our recombinant antibody expression technology, we can generate fully human or animal derived antibodies against virtually any target.
ModiQuest Research has various human and animal derived antibody libraries available. Our animal libraries are derived from naive animals, animals immunized with human tissues or tumors, as well as from animals immunized with various antigens. Our human libraries are derived from:

After selection of the antigen specific antibody with the requested properties, the cDNA sequence can be cloned in our proprietary eukaryotic antibody expression vector set to generate recombinant monoclonal antibodies. Antibodies of various species and isotypes can be generated.
B-cell selection and cloning
For species other than mice and rats (e.g. rabbits, lamas, and humans), the antibody-encoding cDNA from antigen-specific B-cells can be cloned and expressed in a recombinant antibody format.

Antibodies can be produced in continuous culture, following stable cell line generation or batch-wise from transiently transfected mammalian cells.
Antibody sequencing
Antibody encoding cDNA obtained via phage display technology or provided by the client can be cloned and expressed in a recombinant antibody system.

Hybridoma cDNA cloning
ModiQuest Research provides the cloning of antibody-coding cDNA from existing hybridomas into various antibody formats (e.g. human or mouse chimeras). This approach allows for the rescue of valuable antibodies from poorly expressing hybridoma clones. ModiQuest Research has a large set of antibody expression vectors allowing the expression of various human and mouse antibody isotypes in CHO, HEK-293 cells. Antibodies can be produced in continuous culture, following stable cell line generation or batch-wise from transiently transfected mammalian cells.
Stable antibody producing mammalian cell lines
ModiQuest Research provides the generation of stable, high-level antibody producing cell lines. Various mammalian cell lines are used (e.g. CHO, HEK-293, SP/2.0, and NS0).
We use nucleofection and high throughput ClonePix-FL selection technologies in combination with our proprietary vector system. Our approach is fast, efficient, and improves overall throughput.
Transient protein production
ModiQuest Research provides quick and cost-effective transient antibody production. Trial-batches of a given antibody can be produced in transiently transfected mammalian cell culture. The recombinant antibodies are FPLC-purified from the conditioned culture medium.

Novel Therapeutics
